Article Snippet: Background: intravenous immunoglobulin (iViG) is used to provide clinical benefit in a variety of diseases, including autoimmune diseases such as immune thrombocytopenic purpura (itp), neurological disorders, and a plethora of other disorders. the mechanism of iViG effect in itp remains uncertain and in other disorders is unknown.. We have identified a novel mechanism of iViG effect using a mouse model of itp which has allowed a theory to be put forth as to a general mechanism of iViG effect in other disorders.. Methods: cD or balb/c mice were treated with a daily, escalating dose of anti-platelet antibody (anti-cD4 ) to induce itp and maintain platelet nadir over time. iViG ( -2 g/kg) was given by intraperitoneal injection on day 2. platelet counts were monitored by flow cytometry. reticulated platelets were determined by acridine orange staining. thrombopoietic cytokines, thrombopoietin (tpo), interleukin-6 (il-6) and ilwere measured by elisa. total body irradiation (tbi) was given as a single, sublethal dose of 2-4Gy on day 0.
